چکیده

The present study was aimed at examining the distribution and abundance of glycerol dialkyl glycerol tetraethers (GDGTs) of archaeal and bacterial origin in peat samples from surface and deep (ca. 50 cm) horizons of a peat bog in the Jura Mountains (northeastern France). Two principal types of GDGTs are present: extractable GDGTs, recoverable by solvent extraction, and non-extractable GDGTs, linked to the soil matrix. Within the extractable pool, “free” (i.e. core lipids) and “bound” (i.e. intact polar and/or ester-bound lipids) GDGTs can be distinguished. Extractable “free” and “bound” GDGTs were extracted using both accelerated solvent extraction (ASE) and a modified Bligh and Dyer technique. Both methods were shown to allow adequate extraction of “free” archaeal and bacterial GDGTs from soil samples. Both extraction protocols afforded similar relative distributions of archaeal and bacterial GDGTs, although poorer extraction of “bound” GDGTs was observed for ASE relative to Bligh and Dyer. Even though only low amounts of bacterial GDGTs were released after acid hydrolysis of solvent-extracted samples, non-extractable and total extractable GDGTs showed different distribution patterns in some samples. Water table related variations in the abundance of intact archaeal membrane lipids in a Swedish peat bog.

The presence and distribution of isoprenoid glycerol dialkyl glycerol tetraethers (GDGTs), lipids that constitute the membranes of Archaea, have been investigated in a 50-cm long core from a Swedish peat bog. In the acrotelm, the periodically water saturated and thus oxic upper layer of the peat bog, only minor amounts of GDGTs were found. These amounts increase considerably in the catotelm, th...

Distribution of branched glycerol dialkyl glycerol tetraethers in surface soils of the Qinghai–Tibetan Plateau: implications of brGDGTs-based proxies in cold and dry regions

The methylation index of branched tetraethers (MBT) and cyclization ratio of branched tetraethers (CBT) based on the distribution of branched glycerol dialkyl glycerol tetraethers (brGDGT) are useful proxies for the reconstruction of mean annual air temperature (MAT) and soil pH. Recently, a series of 6-methyl brGDGTs were identified which were previously co-eluted with 5-methyl brGDGTs. Howeve...

In situ production of branched glycerol dialkyl glycerol tetraethers in a great basin hot spring (USA)

Branched glycerol dialkyl glycerol tetraethers (bGDGTs) are predominantly found in soils and peat bogs. In this study, we analyzed core (C)-bGDGTs after hydrolysis of polar fractions using liquid chromatography-atmospheric pressure chemical ionization-mass spectrometry and analyzed intact P-bGDGTs using total lipid extract (TLE) without hydrolysis by liquid chromatography-electrospray ionizatio...

A climatic chamber experiment to test the short term effect of increasing temperature on branched GDGT distribution in Sphagnum peat

Branched glycerol dialkyl glycerol tetraethers (br GDGTs) are membrane lipids produced by unknown Bacteria and are being increasingly used as temperature proxies. Nevertheless, the direct effect of temperature on br GDGT distributions has been rarely evaluated. In this study, the impact of increasing temperature on brGDGT distributions and the speed of adaptation of br GDGT source microorganism...
